The plot. Without the plot, there wouldn't be a story to begin with!
The characters. Larger-than-life characters make a story seem more real.
The setting. Where the story is set has a great impact on how it is read.
Perspective. The perspective that a story is in changes how it is understood.

Personally, I think that all the above is important. But personally, I never start writing a story without having a "Wow, this is different!" idea in my head. I just can't write cliched stuff, I just have to get a different idea.
